The University of Adelaide was granted 150 hectares of land in Bedford Park to construct a second campus in 1960 when Flinders University was founded. Planning began in 1961, and construction commenced in 1962.

Australian Labor Party (ALP) won the 1965 State election and chose to turn the Bedford Park campus into an independent university. As a result, the University Act of 1966 was enacted on July 1st, 1966, to formally establish Flinders University after British cartographer and navigator Matthew Flinders. There were 400 pupils and 90 faculty on the first day of school in 1966. Peter Karmel was named the university's first vice-chancellor, and Sir Mark Mitchell was named its first chancellor.

Multiple campuses of Flinders University may be found in southern Australia. The university's main campus, Bedford Park, is located 12 kilometres outside of Adelaide's city centre. It provides a wide range of academic resources, including a large library, continuous Wi-Fi, and computer laboratories. It also has a cricket pitch, football, soccer, baseball grounds, weight rooms, cardio rooms, and well-equipped gyms. In addition, Adelaide's Victoria Square campus, right in the heart of town, is another great place for students to learn and develop.

Tonsley is another location for Flinders University. Additionally, it has several external teaching sites in South Australia, Victoria, and the Northern Territory. There are multiple graduate and postgraduate research programs at Flinders University and many undergraduate and graduate degree programs in a wide range of disciplines, including business and creative arts and engineering and environmental science. Courses are also held in Singapore, Hong Kong, Malaysia, and China.
